You can only use or unlock the touch pad feature of your Asus X54C if you have installed the compatible driver software. If you do not have the driver, you can download it here:
1. Visit
http://support.asus.com/download.aspx?SLanguage=en&p=3&m=X54C
2. Select your Operating System
3. Click the TouchPad link
4. Finally, click the available download link
5. Install your driver software and restart your computer

What you will have to do to enable the touchpad is to press FN and F9 at the same time.
Also you can try removing your mouse device.
You can also go in device manager click on the mouse and property. There you will see a tab where you can enable to use the mouse while a mouse device is still connected.
