first of all the better quality of the input source makes the biggest impact of the picture quality. you always want to match the LCD panels resolution with the input so thee is as little conversion to match the panel (least artifacts or unwanted errors).
In the video section find the contrast and set it for 90%-95% (this controls the bright sections of the picture), the brightness is usually centered (this controls the dark sections of the picture), the color should be around 40% (50% is too much like cartoons), tint or hue is centered, the sharpness is 20% to minimize the hard edges (deemphasizes the edges of changes), and finally the back-light should e set to around 50% according to the lighting in the room.
There are many other settings in the advanced section, the rule of thumb would be to center them and play with them to find your favorite since this all is a matter of taste.
